A New Era of Interferon Studying (#211)
We found a new interferon with extraordinary higher antivirus and antitumor effect than that of the regular interferon (IFNα-2) 1. Therefore, it was named as Super Interferon-I (sIFN-I) 1. Here we do like to emphasize my talking on its super antitumor effect rather than its antiviral effect.
sIFN-I has the direct antitumor for solid tumor by the reduction of tumor size, prevention of recurrence and can completely cure some lung cancer and other cancer by the combined use of sIFN-I with chemotherapy. How to explain the high antitumor effect? We found the spatial structural change of IFN-I with IFNα-2b and intergen of Amgen. We derived our IFN-I from infergen by change the amino coden but without change its amino acid property i.e without change the proteins primary structure. According to the theory of Nobel Prize winner Anfinsen that “the protein primary structure determine its spatial structure”, now we have not change its primary structure, but its spatial structure be changed, therefore the Anfinsen theory should make some modification. The spatial structure was located on its receptor binding region, therefore, the binding affinity and specificity of IFN-I have been significantly changed which makes its downstream genes and events significantly changed. For example, the level of the important interferon action mediator 2’-5’ OAS enzyme increased and has two peaks, this may contribute the high biological effect. In addition to 2’-5’ OAS enzyme, we think there should have more downstream genes changed, thus we have carried out the gene expression and microRNA array and found dramatic change. We also performed the protein and protein interaction and find many binding site of IFN-I on member receptor protein such as C5AR1 and OGF which need further studying.
